Ex Vivo Metrics™: How Drug Studies in Reanimated Human Organs Could Revitalize the Drug Development Process

An editorial by Gerald Curtis, PhD

Fallout from the catastrophic phase I clinical trial of TeGenero’s monoclonal antibody TGN1412 in March 2006,1 in which 6 volunteers suffered life-threatening “cytokine storms,” includes newly tightened regulations in the United Kingdom and renewed concern worldwide about first-in-human trials, particularly for compounds with novel targets or mechanisms of action.

New Service for RNA Isolation and Preparation from Whole Blood
Agencourt Bioscience, NuGEN Technologies, and Affymetrix Clinical Services Laboratory (ACSL) have launched a joint service for automated RNA isolation and target preparation from whole blood samples collected in PAXgene Blood Tubes. This service supports expression analysis profiling using Affymetrix GeneChip technology for drug discovery and development, including biomarker discovery, screening, and cancer research. This service automates the Agencourt RNAdvance Blood chemistry and NuGEN’s Ovation Whole Blood Solution for RNA amplification and labeling, which utilizes the Agencourt RNAClean Kit and the Agencourt SPRIPlate 96R Magnet Plate. (Agencourt Bioscience is a wholly-owned subsidiary of Beckman Coulter, Inc.) The complete workflow delivers high yields from small RNA samples, while eliminating the need for certain steps, such as globin reduction and column purification. Automation is carried out with Beckman Coulter’s ArrayPLEX application on the Biomek FX Liquid Handling Workstation. For more, see company press release
